Do you mean what allows people to be sad (chemical reactions in the brain)? The question is vague, but sadness and pain do have a purpose which are similar physically and emotionally.

There's no simple way to answer this question. It can be circumstance, it could be emotional, it could be physical. Being sad is not a crime, and frankly sometimes very healthy.

Sadness is sometimes conditioned by a person himself/herself while the other times situations make a person sad. The point is nobody deserves to be sad and we all need to help each and every one to smile.
